Reporter & Expression Cell Lines

Upon developing pathways towards an efficient production of glycosylated human proteins or reproducible high content screening of drugs and potentially toxic compounds, roads have been explored leading from the use of primary cells, primary cell in coculture, stem cells, to tissue slices, and towards artificial organs.

BPS Bioscience have been offering reporter cell lines for a range of pathways that are of interest to toxicology and drug development. All cell lines on offer have been engineered to perform their analytical function or expression of target protein in a stable manner. No cell line is transient. Recommended storage is in liquid nitrogen (or -80°C for transport).

Recent knowledge acquired in cancer research about the position of indoleamine-2.3-dioxygenase and PD-1/PD-L1,PD-L2 as promising checkpoints for therapeutic intervention has led BPS to offer cellular assays for these parameters as well. Details below, list of all cell lines available here.

 

Cell Based Inhibitor Screening: targeting PD-1/PD-L1

Coculture of TCR coupled, PD-1 expressing Jurkat T-Cell  (Reporter) and any target cell line engineered to express TCR/PD-L1 (or PD-L2) may lead to ligation and delivery of a flash signal, if the reporter cell line expresses a responsive element adapted to the target cell line.

BPS Cell line #60535 is able to release a flash signal upon presence of substrate (#60690-1), when ligating with an activating  T-Cell Receptor ligand / PDL-1 or PDL-2 on a target cell. A description of the reporter cell line is available here

Use of neutralizing antibodies against PD1 or PD-L1 is one approach to block ligation and promote T-cell activation in this system.
